DESCRIPTION OF THE OFFER

The actions triggered by the activation of GPCR coupled to Gq are key to the development of various pathologies. Classically, Gαq activates PLCβ, which leads to PKC activation and Ca2 + mobilization. However, numerous evidences show the existence of alternative effectors involved in the different functions of Gαq. We have recently described a new effector region in Gαq capable of interacting with proteins that contain PB1 domains. In addition, our group has identified other proteins with PB1 domains such as p62, which also interact with Gq. It should be noted that p62 is a key signaling node in autophagy / mitophagy processes and in redox homeostasis. The objective of this project will focus on the study of the impact of these new interactions on the control mediated by Gq-GPCR of the signaling pathways involved in cell death, different types of autophagy and oxidative stress in different cell models, as well as in determine the extracellular signals responsible for the activation of the Gαq complexes involved in these processes. The identification of new signaling pathways that relate Gαq with autophagy modulation and / or with redox homeostasis will provide a better knowledge of GPCR activation coupled to Gq under pathological conditions and will allow the design of more specific drugs.
