Characterization of Φ29 p5.5, a new viral protein that may be involved in protein- primed genome replication.
As a result of our recent RNAseq transcriptome analysis of Φ29 infected and non-infected Bacillus subtilis (Mojardin and Salas, 2016), we identified a transcript that would encode of a novel phage protein, p5.5. The analysis of its sequence by bioinformatic tools suggested that it could be a DNA/RNA binding protein. Thus, we will attempt to ascertain its actual existence and possible biological role during the viral infection.
Then, we will clone the p5.5 ORF in order to overexpress and purify the recombinant protein from E. coli. This would allow us to analyze its DNA/RNA binding capacity and eventually a possible effect in the viral DNA replication in our standard protocols of Φ29 TP-DNA replication and amplification in vitro.
